免疫细胞培养专用细胞因子

巨噬细胞(Macrophage)通常被认为是由骨髓中的造血干细胞到髓系祖细胞到单核细胞发育而来,迁移到组织后分化为组织特异性巨噬细胞,它存在两个不同的亚群:M1型(经典激活)和M2型(替代激活)。M1型巨噬细胞具有促炎作用,能被脂多糖(LPS)单独或与Th1细胞因子(如IFN-γ,GM-CSF)联合极化,并产生促炎细胞因子,如IL-1β、IL-6、IL-12、IL-23和TNF-α;M2型巨噬细胞具有抗炎和免疫调节作用,能被Th2细胞因子IL-4和IL-13极化,产生IL-10和TGF-β等抗炎细胞因子。M1/M2巨噬细胞的平衡极化对于维系机体健康至关重要。

Description
Mouse bone marrow cells were taken and cultured in a cell culturedish for 3 hours. Following this, the cells were transferred to a bacteria culture dish and cultured inmedium containing 20 ng/mL M-CSF, The medium was changed every 3 days, and after 7 days, the cells were matured, Flow cytometrywas employed to assess changes in macrophage markers, revealing a positive rate of macrophages as high as 98%. Additionally, results from phagocytosis experiments indicated that the induced macrophages exhibited strong phagocytic capability.

Description
1、Extract the bone marrow cells of a 8-week-old male C57 mouse, stimulate the cells with mouse CSF-1 (Cat: RP01216) at a final concentration of 20 ng/mL for one week (change the culture mediumevery day). Collect the cells, and detect the result of macrophage induction by flow cytometry.

2.Stimulate the M0 cells with mouse IL-4 (Cat. RP01161) at a final concentration of 40 ng/mL. Then collect the cells detect the result of M2 polarization by flow cytometry.

操作步骤

细胞因子复溶及保存操作收起
  • 1. 未添加载体蛋白的产品,如RP01216SRP01161等操作如下:
    使用含0.1% BSA或者10% FBS的无菌PBS将重组M-CSF细胞因子复溶至100 μg/mL(10 μg因子加100 μL PBS,100 μg因子加1 mL PBS,以此类推),将复溶后的M-CSF母液按10 μL/管进行分装,冻存在-20℃以下,随取随用,避免反复冻融。
  • 2. 本身已添加了载体蛋白的产品,如RP01216SCRP01161C操作如下:
    使用无菌PBS将重组M-CSF细胞因子复溶至100 μg/mL,无需额外添加保护剂(10 μg因子加100 μL PBS,100 μg因子加1 mL PBS,以此类推),将复溶后的M-CSF母液按10 μL/管进行分装,冻存在-20℃以下,随取随用,避免反复冻融。
  • 3. 含0.1%BSA的PBS配制方法如下:称取10 mg BSA溶解在无菌PBS中,定容至10 mL,用0.22 μm的滤膜过滤除菌后使用。
获取小鼠骨髓细胞收起
  • 1. 取C57BL/6J雄性小鼠(6-8周龄)颈椎脱臼法处死,手术取出所有股骨和胫骨,并用剪刀和镊子将骨周围的肌肉组织尽量去除干净,不要损伤到骨;
  • 2. 将骨移至超净台内,并用盛有70%酒精的无菌培养皿浸泡2-5 min,以消毒灭菌,然后用无菌PBS(含1% P/S)洗2次;
  • 3. 将骨移入另一个盛有PBS(含1% P/S)或RPMI 1640(含1% P/S)的新培养皿中,用剪刀剪去骨两端,再用注射器抽取PBS(含1%P/S)或RPMI 1640(含1% P/S),针头分别从骨两端插入骨髓腔,反复冲洗出骨髓至培养皿中,直至骨完全变白;
  • 4. 收集骨髓细胞悬液,用200目尼龙网滤去小碎片和肌肉组织;
  • 5. 滤过液400g离心5 min,弃上清;
  • 6. 加入5mL 红细胞裂解液吹打混匀,室温裂解5min后,400g离心5 min(如果细胞沉淀仍有较多红细胞,可重复裂解一次);
  • 7. 使用完全培养基(α-MEM+10%FBS(热灭活)+1% P/S)重悬细胞,即获得小鼠骨髓细胞,一般一只小鼠大约可获得2×107-4×107个骨髓细胞;
  • 注:红细胞裂解时最长不超过10 min,以免对有核细胞造成损伤。
M0巨噬细胞诱导收起
  • 1. 使用完全培养基(α-MEM+10%FBS(热灭活)+1% P/S)将细胞培养过夜,以去除贴壁较快的其他杂细胞如纤维细胞等等;收集未贴壁细胞,调整细胞浓度为2×106-3×106/ mL,铺板培养(10 cm未TC处理的细胞培养皿,每皿10 mL细胞悬液),在完全培养基(α-MEM+10%FBS(灭活)+1%P/S)中添加50 ng/mL M-CSF诱导向巨噬细胞成熟;每隔1天换液一次;
  • 2. 培养4-6天分化为成熟巨噬细胞(贴壁),倒置显微镜观察细胞形态,同时选择F4/80+ CD11b+双阳表达作为鉴定巨噬细胞成熟的标志。
  • 注:(1)血清热灭活处理方法,56℃处理30 min。目的是使血清中的补体失活而不影响其他的有助于细胞生长的成分,避免补体对培养的巨噬细胞不必要的激活或者破坏作用;
    (2)巨噬细胞的生长和增殖依赖于M-CSF,如果诱导过程中发现细胞漂了,可能的原因之一是M-CSF耗尽,需要补加因子;
    (3)使用未TC处理的培养皿/培养板进行培养,目的是便于将贴壁细胞吹洗下来,避免用胰酶消化影响细胞活率。
M1/M2型巨噬细胞极化收起
  • 1. 将成熟的M0巨噬细胞用无菌PBS轻轻吹洗离心2遍后,重新调整细胞密度5×105/孔(未TC处理的6孔板)并铺板,添加相关细胞因子,诱导其向M1、M2型巨噬细胞的极化:
    M1极化因子:IFN-γ(50 ng/mL)+ LPS (100 ng/mL)
    M2极化因子:IL-4 (核心因子, 20 ng/mL),IL-13(可选,20-50 ng/mL), IL-10(可选,20-50 ng/mL)
  • 2. 刺激24-48 h后,用流式检测M0(F4/80+, CD11b+)和极化后M1(CD86+, CD80+等)、M2(CD206+,Arg-1+等)标志蛋白的表达
    对极化巨噬细胞进行形态学观察:巨噬细胞极化后形态发生变化,M1巨噬细胞通常呈现出更圆或不规则的形态,M2巨噬细胞倾向于呈现细长的形态,有时会形成纺锤形的细胞。
巨噬细胞流式鉴定操作收起
  • 1. 将细胞重悬于PBS中,调整细胞浓度至1×107/mL。
  • 2. 每管加入100 μL细胞悬液(约1×106细胞),加入相应的的荧光标记抗体,并设置相应的对照(根据抗体说明书调整),经典鉴定方案如下:
    M0:F4/80+,CD11b+
    M1:F4/80+,CD11b+,CD80+/CD86+, CD206-
    M2:F4/80+,CD11b+,CD80-/CD86-, CD206+
  • 3. 避光孵育15 - 30分钟,4℃。
  • 4. 加入2 mL PBS洗涤细胞,1500 rpm离心5分钟,弃上清。
  • 5. 重复洗涤一次,重悬细胞于200 μL PBS中,上机检测。
  • 注:CD68和CD206为跨膜抗原,需要确定相应的克隆号为识别胞外或者胞内区域,流式染色需要做固定破膜胞内染色。

树突状细胞(Dendritic cell, DC)是一类来源于骨髓造血系统的异质性细胞,可由髓系祖细胞和淋巴祖细胞发育而来,是目前发现的功能最强的抗原呈递细胞(APC)。DC细胞能在天然免疫中识别病原体,在适应性免疫中通过MHC分子呈递抗原肽来诱导Naïve T细胞激活和分化,是先天免疫和适应性免疫之间的主要桥梁。在常规的DC诱导培养中,GM-CSF、IL-4和TNF-α被认为是最重要的三大细胞因子,其中GM-CSF促进DC生成;IL-4通过抑制DC前体细胞向巨噬细胞分化,而分化为未成熟的DC;加入TNF-α后,DC前体细胞即向成熟表型DC分化。

自然杀伤细胞(Natural killer cell,NK细胞)来源于骨髓造血干细胞,属于先天免疫系统的核心细胞,由共同淋巴祖细胞发育而来,也是除T细胞和B细胞之外的第三大类淋巴细胞。NK细胞主要分布于外周血、肝脏和脾脏,通过激活和抑制信号的控制来识别和杀死靶细胞。由于具有无MHC限制的自然杀伤活性、通过分泌IFN-γ等细胞因子和趋化因子参与免疫调节、能介导抗体依赖细胞介导的细胞毒性作用(ADCC),NK细胞常被用于开发肿瘤免疫细胞疗法。在体外培养过程中常用IL-2、IL-12、IL-15、IL-21等细胞因子刺激NK细胞,提高其活性。

T细胞是适应性免疫能力的重要组成部分,可对病原体启动细胞介导的免疫应答,大致分为辅助性T细胞(CD4+T细胞)和细胞毒性T细胞(CD8+T细胞)。CD4+T细胞包含Th1、Th2、Th9、Th17、Th22、Treg和Tfh等不同的亚群,通过分泌细胞因子产生炎症或激活其他免疫细胞。CD8+T细胞按照分泌的细胞因子的不同分为Tc1,Tc2,Tc17, Tc9和Tc22等亚群,可直接杀死被病原体感染或癌变的细胞。每种T细胞亚群都由一组特定的细胞因子和转录因子活化,这些细胞因子在T细胞增殖、分化的培养过程中发挥了重要作用。

B细胞是一种能够对病原体启动抗体介导的免疫应答的适应性免疫细胞,可生成高亲和力抗体、形成免疫记忆、充当抗原呈递细胞,并分泌细胞因子。骨髓中的B细胞源自造血干细胞,由其分化为共同淋巴祖细胞,随之生成自然杀伤细胞、T淋巴细胞和B淋巴细胞。在B细胞的发育过程中,有多种细胞因子会影响其生长、存活和抗体类型转换,例如IFN-α和IFN-β主要参与B细胞的生成和选择,而IL-4和IL-6则影响B细胞优先分泌特定类别的抗体。

B细胞培养常用细胞因子

Description
B cells were cultured using mouse IL-4 (1 ng/mL, Cat. RP01161) for 4 days. Obvious cluster-like growing cells could be observed, indicating that IL-4 provides the necessary signals for the proliferation and activation of B cells.
