Recombinant Mouse IL-4 Protein was determined by SDS-PAGE under reducing conditions with Coomassie Blue.

Recombinant Mouse IL-4 Protein is greater than 95% as determined by SEC-HPLC.

Recombinant Mouse IL-4 promote the proliferation of MC/9 2 mouse mast cells. The ED50 for this effect is 2.8-11.3 pg/mL, corresponding to a specific activity of 8.85×107~3.57×108 units/mg.

Recombinant Mouse IL4 stimulates cell proliferation of HT-2 mouse T cells. The ED50 for this effect is 1.18-4.70 ng/mL, corresponding to a specific activity of 2.13×105~8.51×106 units/mg.

描述
Recombinant Mouse IL-4 Protein is produced by HEK293 cells expression system. The target protein is expressed with sequence (His23-Ser140) of mouse IL-4 (Accession #NP_067258.1) fused with a 6×His tag at the C-terminus.
储存
Store at -20℃.Store the lyophilized protein at -20℃ to -80 ℃ up to 1 year from the date of receipt.
After reconstitution, the protein solution is stable at -20℃ for 3 months, at 2-8℃ for up to 1 week.未开盖的干粉蛋白在 -20°C至-80°C可保存12个月;
复溶之后,蛋白溶液在-20°C及以下可保存3个月,在2-8℃可保存1周。
生物活性
1.Measured in a cell proliferation assay using MC/9 2 mouse mast cells. The ED50 for this effect is 2.8-11.3 pg/mL, corresponding to a specific activity of 8.85×107~3.57×108 units/mg.
2.Measured in a cell proliferation assay using HT-2 mouse T cells. The ED50 for this effect is 0.21-0.85 ng/mL, corresponding to a specific activity of 1.18×106~4.76×106 units/mg.
复溶
Centrifuge the vial before opening. Reconstitute to a concentration of 0.1-0.5 mg/mL in sterile distilled water. Avoid vortex or vigorously pipetting the protein. For long term storage, it is recommended to add a carrier protein or stablizer (e.g. 0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose), and aliquot the reconstituted protein solution to minimize free-thaw cycles.收到重组蛋白产品之后请检查蛋白冻干粉末是否贴于瓶底,如果粉末浮起,开盖之前请先低温离心。将蛋白用说明书中指定的缓冲液复溶至0.1-0.5 mg/mL(请注意蛋白复溶浓度不能低于0.1 mg/mL),室温平衡5-10 min保证充分溶解,复溶过程中请不要剧烈涡旋及吹打蛋白溶液。如需长期储存,建议复溶时添加载体蛋白或者稳定剂(如0.1% BSA, 5% HSA, 10% FBS 或者 5% 海藻糖),同时将复溶后的蛋白溶液按照需求进行分装,储存于-20°C至-80°C,随取随用,避免反复冻融。

1、Extract the bone marrow cells of a 8-week-old male C57 mouse, stimulate the cells with mouse CSF-1 (Cat. RP01216) at a final concentration of 20 ng/mL for one week (change the culture medium every day). Collect the cells, and detect the result of macrophage induction by flow cytometry. 2、Stimulate the M0 cells with mouse IL-4 (Cat. RP01161) at a final concentration of 40 ng/mL. Then collect the cells detect the result of M2 polarization by flow cytometry.

In vitro induction culture of mouse bone marrow derived dendritic cells (BMDC): C57BL/6 mouse bone marrow cells from femur and tibia were induced with IL-4 (10 ng/mL, Cat. RP01161) and GM-CSF (20 ng/mL, Cat. RP01206) to differentiate into BMDC. DCs were labeled with CD11c, and DCs maturation was detected by CD80 and CD86 at 7 days after induction. Flow cytometry result showed that the proportion of cells expressed CD11c was as high as 89.4%, and more than half of the cells expressed CD80 and CD86, indicating BMDC was successfully induced.

On the sixth day of culture, mouse macrophages were added with: IL-10 (10 ng/mL, Cat. RP01465) + IL-4 (20 ng/mL, Cat. RP01161) to induce M2 macrophage polarization, and IFN-γ (50 ng /mL, Cat. RP01070) + LPS induced the polarization of M1 macrophages for two days. The changes in relevant markers were detected by flow cytometry, and the results showed that the induction was successful.

Interleukin-4, also known as IL4, is a secreted protein that belongs to the IL-4 / IL-13 family. Interleukin-4 / IL4 has many biological roles, including the stimulation of activated B-cell and T-cell proliferation. It enhances both secretion and cell surface expression of IgE and IgG1. Interleukin-4 / IL4 also regulates the expression of the low-affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes. Interleukin-4 is essential for the switching of B cells to IgE antibody production and the maturation of T helper (Th) cells toward the Th2 phenotype.
