Under the stimulation of various ligands such as IL-2, GM-CSF, growth hormone and prolactin, Stat5 is activated. The activation of Stat5 requires Tyr694 phosphorylation. Under the stimulation of erythropoietin, Src can mediate this phosphorylation. Stat5 is constitutively activated in certain types of leukemia cells. Phosphorylated Stat5 was found in some endothelial cells treated with IL-3, indicating its association with angiogenesis and cell motility. Stat5a and Stat5b are independently regulated and activated in different cell types. For example, interferon treatment mainly activates Stat5a in U-937 cells and Stat5b in Hela cells.
